Peripheral T-Cell Lymphomas of the T Follicular Helper Type: Clinical, Pathological, and Genetic Attributes

نویسندگان

چکیده

Follicular helper T-cell (TFH) lymphomas comprise a unique group of that represent neoplastic proliferations follicular T-cells and share genetic, immunophenotypic, morphologic, clinical features. Angioimmunoblastic lymphoma (AITL) is the prototypical TFH lymphoma; in addition, 2017 revised World Health Organization (WHO) 4th edition recognizes two other subtypes: (FTCL) nodal peripheral with T phenotype (PTCL-TFH). This review discusses morphologic spectrum, immunophenotype, diagnostic mimics/pitfalls, genetic attributes this category lymphomas.

Peripheral T-cell lymphomas with a follicular growth pattern are derived from follicular helper T cells (TFH) and may show overlapping features with angioimmunoblastic T-cell lymphomas.

Rare cases of peripheral T-cell lymphomas with follicular growth pattern (PTCL-F) have been recently reported, and their association with t(5;9)(q33;q22) involving ITK and SYK has been suggested. However, the clinicopathologic aspects of PTCL-F are poorly described and the normal cell counterpart of this subgroup of lymphoma is still unknown. Therefore, we analyzed the pathologic, phenotypic, a...

Immunophenotyping of Nodal Peripheral T-cell Lymphomas and its Association with Epstein-Barr Virus

Background: Immunophenotyping in the rare group of nodal Peripheral T-cell Lymphomas (PTCL) exposes interesting features such as T-cell marker downregulation and paradoxically, the presence of reactive, clustered large-sized CD20 positive B-cells (B-cell proliferation). Epstein-Barr virus (EBV) has been suggested as a putative etiology in pathogenesis of B-cell lymphoma. We aimed to review the ...

Peripheral T-cell lymphomas.

The development of T cells from stem (progenitor) cells to effector cells results from a two-wave process of proliferation and differentiation. The cells of the first differentiation wave are the precursor T cells, and those of the second differentiation wave are peripheral T cells. In the first differentiation wave, resting/circulating naive antigen-reactive T lymphocytes are produced which di...

Follicular helper T cell signature in type 1 diabetes.

The strong genetic association between particular HLA alleles and type 1 diabetes (T1D) indicates a key role for CD4+ T cells in disease; however, the differentiation state of the responsible T cells is unclear. T cell differentiation originally was considered a dichotomy between Th1 and Th2 cells, with Th1 cells deemed culpable for autoimmune islet destruction. Now, multiple additional T cell ...

T follicular helper cells, T follicular regulatory cells and autoimmunity.

CD4(+)T follicular helper (Tfh) cells are recognized as a distinct T-cell subset, which provides help for germinal center (GC) formation, B-cell development and affinity maturation, and immunoglobulin class switching, as an indispensable part of adaptive immunity. Tfh cell differentiation depends on various factors including cell-surface molecule interactions, extracellular cytokines and multip...
